Biography:

Dr. Ernstoff received his BA in the history of art from Emory University in 1974 and his M.D. from New York University of Medicine in 1978. After completing internal medicine training at the Bronx Municipal Hospital Center and the Hospital of Albert Einstein College of Medicine, he continued his training in Medical Oncology at Yale University, where he stayed on faculty as an Assistant Professor of Medicine and Director of the Office of Clinical Research of the Yale Comprehensive Cancer Center until 1986. In 1986, Dr. Ernstoff joined the faculty at the University of Pittsburgh and the Pittsburgh Cancer Institute, where he was an Associate Professor of Medicine and Surgery, Director of the Hematology/Oncology Fellowship Program, and the Medical Director of Genitourinary Cancer Group until 1991 at which time he joined the faculty of the DHMC. Dr. Ernstoff is currently an oncologist in the field of GU and Melanoma.

Research Interests:

Dr. Ernstoff's laboratory has been involved with developing clinical use of cytokines in the treatment of metastatic renal cell carcinoma or melanoma. Correlative laboratory studies suggest that T cell mediated pathways may be triggered by the interferons causing tumor regression. Further studies exploiting this observation include trials employing tumor vaccines and sequential combination of cytokines. The development of tumor specific vaccines for the treatment of renal cell carcinoma and prostate cancer are currently being explored in the laboratory and clinic. Cytokine modulation of vaccine response is currently being explored using autologous renal cell carcinoma and interferon alpha combined with interferon gamma in patients with metastatic renal cell carcinoma. Preliminary studies suggest that cytolytic T cell pathways may be activated through this approach. In a related area, the laboratory is attempting to identify tumor-specific immunodominant peptides that exist on prostate carcinoma. Immunodominant peptides are identified using the Tsites program, which predicts for 8-15 amino acid sequences that bind to the major groove of the MHC molecule. To enhance peptide presentation to monocytes, immunodominant sequences are conjugated to a monoclonal antibody to the Fc-gamma RI receptor.
